Doctor Otto Gunther Octavius is a genius but highly egotistical scientist who defected from Oscorp while working on gamma-fusion designs. In his efforts for fame and recognition, he began inventing and selling advanced weaponry to criminals in New York City, such as Butane, Speed Demon, Maria Vasquez, Unicorn and Scorpion. This attracted his former colleague Norman Osborn, who then had Octavius arrested for his dealings and violations of the Sokovia Accords.

Otto Octavius' design is heavily based on John Romita Sr.'s design of Doctor Octopus from the comics, while his glasses are based on the ones seen in the Spider-Man (1994) animated series.
- Unused concept art of Spider-Man: No Way Home shows Alfred Molina's Doctor Octopus wearing a variation of the green and yellow lab suit.
The symbol on Otto Octavius' lab coat, yellow lab glove and all his inventions is a combination of two octagons, two "O" letters representing his initials, an infinity symbol (∞), and the number 8 being tilted sideways to represent being Doctor Octopus.
