The Ancient One was the Sorcerer Supreme as well as the leader of the Masters of the Mystic Arts. For centuries, the Ancient One protected Earth from mystical threats, and dedicated herself to helping others in need by educating them in the magical arts. The Ancient One encountered Doctor Strange and accepted him as her student before being murdered, after which Strange succeeded in her role. However, Strange became bent on trying to resurrect his girlfriend Christine Palmer by undoing her death through time travel, which threatened the stability of reality itself. In a last effort, the Ancient One caused a paradox to create an alternate Doctor Strange to defeat his rogue counterpart, though they had ultimately failed.
The Ancient One accepted Stephen Strange into the Masters of the Mystic Arts as a disciple when he was searching for answers after the death of Christine Palmer. Under her mentorship, Strange learned how to use the Eldritch magic which made him a powerful sorcerer. However, Strange's curiosity in the Eye of Agamotto deeply concerned the Ancient One and Wong, who warned Strange about the possible consequences of tampering with the matter of time.
The Ancient One and the Masters of the Mystic Arts were confronted by beings who were seeking to summon Dormammu onto the Earth plane. Eventually, Ancient One was severely wounded and the doctors at the Metro-General Hospital failed to save her from death. With the Ancient One passing away, she passed the title of the Sorcerer Supreme to Strange, and he succeeded in saving Earth from Dormammu.[1]

Ancient One explains the Absolute Point in time to Doctor Strange
"Stephen, this path only leads to darkness and the end of this reality. Please do not do this, or I will be forced to stop you."
In an attempt to save Christine Palmer from her death, Doctor Strange used the Eye of Agamotto to travel back in time, however, her death was inevitable despite his efforts. The Ancient One from that time approached devastated Strange and told him that Palmer's death was an Absolute Point in time, an unchangeable event that was supposed to be the beginning of Strange's journey as the Master of Mystic Arts. Strange begged the Ancient One to help him bring Palmer back, however, she replied that the Absolute Point can not be reversed even by the greatest sorcerers, but Strange remained unconvinced.
The Ancient One warned Strange that the path he had chosen would lead to the end of their reality, however, he remained certain that he only needed more power to fix everything. Noticing that Strange kept ignoring her warnings, the Ancient One decided to stop him by force and when Strange attempted to escape, she cast a spell on him. Using the Dark Dimension power, the Ancient One split the timeline into two separate parts; while the original Strange was isolated, an another version that was able to overcome Palmer's death was left in the timeline to live.[1]
"You cannot save her, Stephen. The resulting temporal paradox will destroy this universe. Our time is running out. I fear the only sorcerer strong enough to stop Doctor Strange is... you, Doctor Strange."
However, Doctor Strange refused to give up and started to study at the Lost Library of Cagliostro, gaining more power to reverse the Absolute Point in time and save Christine Palmer. An alternate Doctor Strange was visited by the psychic impression of the Ancient One sent through a splinter in reality. The Ancient One told Strange about his counterpart's plans to save Palmer which would result in the destruction of their reality which prompted her into splitting the timeline. The Ancient One explained to Strange how devastating the consequences would be and asked him to stop his alternate self from reversing the Absolute Point, shortly before vanishing.[1]
Magic: As the Sorcerer Supreme, and the leader and the most powerful member of the Masters of the Mystic Arts, the Ancient One was a tremendously powerful and skilled master sorceress, possessing immense magical power and absolute mastery and knowledge of the mystic arts.
Immortality: The Dark Dimension energy suppressed the Ancient One's aging process, prolonging her lifespan greatly.
Timeline Manipulation: The Ancient One was able to split her universe main timeline into two alternate timelines that coexists simultaneously in the same universe. She used that when she split the timeline and created a temporal paradox where twoversions of Doctor Strange coexisted in the same timeline in different moments.

Ancient One was able to send a psychic impression of herself from the afterlife through a splinter in reality to tell Doctor Strange about his counterpart's plan to save Christine Palmer.
Metro-General Hospital: The Ancient One was brought to the Metro-General Hospital after being impaled. The doctors there were not able to save her life, and she passed away at the hospital.
