Unfortunately, it's very unclear.
Evidence Strange was Sorcerer Supreme from January 2017 to May 2018:
In January 2018 in What If...?, he is Sorcerer Supreme in an otherwise very similar universe up to that point.
Peter thought Strange was Sorcerer Supreme, as if he had good reason to believe that.
Strange's only correction to Peter saying that was Wong being Sorcerer Supreme instead of him because of the Blip, not "No I never was".
Evidence Strange wasn't Sorcerer Supreme from January 2017 to May 2018:
Strange says no in reply to "I thought you were Sorcerer Supreme?", and while he might just be taking Peter's line as purely meaning present day, he says "No" as if it's a flat simple no to the whole thing, that part of his reply somewhat implying he was never Sorcerer Supreme.
He doesn't know about the Book of Vishanti's existence, which the Sorcerer Supreme learns from one of their special books.
While some of Wong's teasing could work fine as "Ha, I'm the Sorcerer Supreme now, not you anymore", some of it doesn't, like the book about the Book of Vishanti, which he's teasing Strange for seemingly never having had the chance to read.
I don't think, even if Strange wasn't, that Wong was, since Strange makes a big point about the Blip being the reason for Wong.
My guess is they haven't worked it out precisely. However, to reconcile it, I imagine from January 2017 to May 2018 there was no Sorcerer Supreme, but during that time Strange was chosen to take over and was training for it or something (hence Peter thinking he was the Sorcerer Supreme, and as close as possible to matching the What If...? universe). Then the Blip happened, so Wong took over (hence Strange feeling Wong got it instead of him on a "technicality" due to the Blip), and Strange had never actually been Sorcerer Supreme (so never read the book, Wong can tease him - and hence the lack of explicit mention to Peter that he was).

@BEJT I like that explanation! It is indeed weird, I always assumed that Strange was the sorcerer supreme between DS1 and IW. I also did find the whole Book of Vishanti thing quite odd when that was placed on the table. Those thoughts lead those things to make more sense.
@ThePunisherMeetsSpidey The source on that alias is No Way Home. That seems like an assumption from an editor, or it's based on Peter referring to him initially as Sorcerer Supreme and it's the weird way the Alias section works that that counts as something someone called him, regardless of its veracity.
In a new interview with Empire (Spoiler Special Podcast, behind a paywall), 40 minutes in, Richie Palmer confirms: "To clarify that, too, Strange was never the Sorcerer Supreme. I think that after the Ancient One's demise in the first movie, I don't think anybody was clamouring for it, let alone Strange."
He then elaborates his own personal interpretation: "I think everybody in that world probably assumed it would be him, he was like, 'I got bigger fish to fry' [...] I think Strange was probably a little after-the-fact like, 'Oh dammit, I could've had that.'"
